Melanotan II is a synthetic peptide that acts as a non-selective agonist of melanocortin receptors. Originally developed as a tanning agent, it earned the nickname "Barbie" peptide due to its ability to bind to MCR1 receptors. Melanotan II is a modified version of Melanotan I, designed with a longer half-life. It is a cyclic lactam analog of α-MSH (melanocyte-stimulating hormone), which is known to regulate skin pigmentation by stimulating the production of melanin in melanocytes.

Melanotan II influences melanogenesis, the process by which melanin is produced, which determines skin, hair, and eye color. Melanin also plays a crucial role in photoprotection by absorbing ultraviolet radiation (UVR), reducing the risk of UV damage. Additionally, melanocytes interact with various systems in the body, including the endocrine, immune, inflammatory, and central nervous systems, and are regulated by external factors like UV radiation and medications.

Unlike Melanotan I, Melanotan II can cross the blood-brain barrier and interacts with five melanocortin receptors (MC1R-MC5R), each with specific tissue expression patterns. The MC3R and MC4R receptors, in particular, are important in regulating feeding behavior and body weight.
